a basic manual has a clutch pedal and gear selector that is mechanically attached to the gearbox.
a basic automatic has a fluid coupling connecting the engine and gearbox (rather than a clutch) and a computer or flywheel controlling the gearchanges. modern automatics can even take driver input on when to shift. this in Audis is called tiptronic. it really isnt faster at shifting than people despite the fact that people say it is. the reason for this is because of the huge ambiguity created by the term flappy paddle gearbox. since automatics can take driver inputs, a way of getting this input can be through paddles behind the wheel. it's just the same as an old automatic that had P, R, N, D, 3, 2, and 1 on the selector column. you still have to deal with the power and response loss of a fluid coupling. if you don't understand what a fluid coupling is, please look here. wwwube/watch?v=leCEmJA0WsI
now the other gearbox types are a bit more complex and typically more expensive.
automated manuals are gearboxes that are connected to the engine with a clutch (like a manual) unlike a manual, a computer controls that clutch. this means that you get the immediacy in response of a manual with the ease of driving of an automatic. now a cople years ago, these systems were just starting to get popular so the programs controlling the gearshifts were not that refined. at high speeds, the gearboxes were very quick but on roads they felt odd. gearshifts would be slow (to try and maintain comfort) but at the same time very hard (since a computer wasnt that good at modulating the clutch). nowdays, these programs have gotten very good and automated manuals feel like a regular automatic with the exception that when you let off the brakes you don't creep forwards.
added to that, companies in an attempt to get more speed out of cars decided to split up the gears over 2 clutches. this is the premise behind a dual clutch automated manual. (mostly referred to as a dual clutch gearbox, the fact that the clutches are controlled by a computer makes them automated manuals). by splitting up the gears, they could shorten the time between shifting and getting a much lower drop in power. This is done by having 1 clutch with the next gear already engaged, all that's required is the switch in clutches to switch gears rather than a clutch activation, gear change then clutch deactivation. This meant that under full acceleration, cars got faster. added with smoother shifting programs, a dual clutch gearbox is the best performing gearbox type you can get in a road car.
now the last gearbox type is a sequential and I won't be going over it since there are numerous variations on them the main premise is that instead of having the ability to manually select whatever gear you want, you can only go up one or down one. this is especially useful in racing where clutchless shifts are popular in order to decrease shifting times and extend gearbox longevity.
now to the audi specific side.
R-Tronic was the original gearbox used in the Audi R8. it was a single clutch automated manual.
S-Tronic is the new dual clutch automated manual in the R8 Plus models. Having personally driven an R8 4.2 V8 R-tronic, I know for a fact that the basic R-tronic was sluggish and really awkward at low speeds. gear changes feel slow and brutal at the same time and for a car of that price and performance it should have had much better response times. Audi released a software update to their R-tronic gearbox that had a great effect on how the car felt around town. (the same software updates that make a dual clutch great in town). unfortunately, shift speeds can't be improved too much because of the single clutch design but the smoothness of the system was drastically improved the new s-tronic, people say that it greatly improves how the car feels around town AND at the same time has a fairly good effect on acceleration and top speed (along with the extra 25 hp that they got from the engine).

I'm a huge Audi fan and I want to know about these two transmission systems. I've looked at some other websites but they go on about this crazy stuff about "dual-clutch" and "automated manual" and stuff. Tiptronic was first developed by porsche and now used by audi. It's basically a "semi-auto" transmission which allows the user manually up or downshift, with some limitations. R-tronic is a sequential manual gearbox like that of bmw's smg. It's really lamborghini's e-gear just renamed for use it the R8. S-tronic is the same as vw's dsg.
